Getting around Tyria is all about missions or running. The run from lions arch to ascalon is not difficult for a runner of which you will find them popping up in LA regularly. Tho the cost is usually a little on the steep side due to the length of the run and not necessarily the difficulty.

Since you seem to want to get an Assassin there, I'd suggest you get a runner. It's a pain in the backside to get past the dwarves who insist on using Pin Down. Without them, you'd probably blast past them no problem, but...

From LA, run north through North Kryta Province, east through Scoundrels Rise, north east though Griffon's Mouth, east then north through Deldrimor Bowl (stopping at Beacons Perch if someone wants to), north east through Anvil Rock(stopping at Ice Tooth Cave if someone wants to), east through Iron Horse Mines, south east through Traveler's Vale(stopping at Yak's Band if someone wants to), south east through Ascalon Foothills, south east again through Diessa Lowlands (stopping at Grendich Courthouse if someone wants to), east and a bit south through The Breach (stopping at Piken Square if someone wants to), then east through Old Ascalon.

Enjoy.

If you take your time, you can do it with Henchies, and the closer to Ascaslon, the easier it gets, due to the diminishing skill lvl of the bad guys.
My lvl 15 Rit had no problems.
