Yes, you can repeat a mission as many times as you want. AFAIK there are no other bonueses to your first question, but I'm not very experienced in PvE.

You get more gold and more exp by completing the missions on master level, which helps skill gaining too by giving more skillpoints with level ups and money to buy the skills. And to some the title is reason enough, including the challenge it usually entails.
