PvP is Player vesres Player, PvE is the story-line part of the game where you fight AI enemies. They're both pretty popular.
There is no "most effective" profession. The thing to do is try different ones and see which ones you enjoy playing. You'll be better at the ones you like to play.
The easiest to play probably Warrior. Most people would say that Mesmer is the toughest, since it requires knowing a lot about what the other professions do to effectively combat them. Which is more fun is a matter of opinion. If you like making lots of noise and flashy effects, then Elementalist would be the way to go. If you like going toe-to-toe wiht your foes, Warrior or Dervish. Try different ones to see what suits YOUR playing style.
You'll be able to get your secondary profession once you reach level 5. For tons of good info on all things GW, check out

It's better to party up, even if it's only with heros and henchmen, you won't get as much XP per kill, but you won't get your butt handed to you every time you set foot out of town. (Be sure to talk to the sunspear scouts by the rez shrines for bounties & double XP.)
Dervish is a front line damage dealer, Paragons are more of a support class.
There's lots of things to learn; collectors, skills, traders, team composition, and skill sets. But it won't take long to get the hang of the game mechanics, get over the initial "What the hell am I doing?" phase, and get leveled up to 20, where the real challenge of the game begins.
The best thing to do as a new player is learn the difference between "noob" and "newbie." Seriously, while in-game, talk to every NPC you see to find out what services & products they have to offer. Also check out guildwiki, you'll find stuff about scams, armors, professions, and all kinds of neat things.
