In terms of where to go, which NPCs to talk to, and which quest lines to complete, what recommendations are there for getting the most value out of the 5-hour limit?

don't need to kill the wurms outside boreal station, one can run from boreal to eye of the north with minimal fighting involve, get the hero's handbook once there. go into Hall of Monument, talk to Gwen, talk to scarying pool, talk to vekk, talk to ogden, come out do the northern allies quest. ie:tracking the norn bear, because its the closest.

i recommend norn tournament. allows you to get Zehtukas horn and the Zhos book for the miniature chick. Just follow the story, you get there quick.

Then decide exactly what you want to do, i.e:

Norn, Asuran or Vangard arc. Norn is easiest, because you start there, but you can leg it to the other territories. Just remember to pick up all the primary quests at The Eye/HoM or you'll encounter gates.

Or maybe take in a dungeon or two? "Sepulchre of Dragrimmar" is close to the Norn outpost Sifhalla, and really short, if you don't insist on fighting everything.

If it were me - I'd follow the Norn arc as far as "Sifhalla" and then head into "Jaga Moraine", do the "Frostmaw's Burrows" dungeon and then squeeze in Sepulchre, if there's still time.

Finally, get your current heroes ready and read up wiki pages relating to any of the above *before* activating the key.

Whatever you decide - good luck!

of course that skill will be and remain locked untill you actually buy EotN after the trial.
Getting Zho's Journal was a very good tip ^^
I thought of buying the Tyrian armor, but that's the same as the one you can get in Factions at Voldo the Exotic.
So if you don't have factions and like the skin, buy that one. Prophecies also has it, but only collectors have max armor, and those can't have insignia's anymore.
(do assassins and ritualists get basic exotic armor in Boreal station? And what do Dervishes and Paragons get there?)

Let's face it, the 5-hour trial is just a teaser to get you to buy the expansion. There isn't really enough time to do anything significant. You could just concentrate on getting particular skills or items, but most of them aren't much use unless you get the expansion - and then you could get them anyway (and they may not be useable until you get EotN.)

So I'd say your best bet is to get to Eye of the North (the outpost) and from there, try to get to a dungeon (or perhaps, just Battle Depths) to see what they are like.
If you don't have Nightfall, do some of the quests that get you heroes so you can try them out too.
