Can you kill Duncan multiple times after killing the 4 bosses or only 1 time? (assuming you don't hand in the quest)

If I'm not mistaken, even if you hand in the quest, you can get it again and kill Duncan without killing the others first.

You're mistaken.
If you hand in the quest, you cannot access Duncan's level until you repeat and kill the other 4 bosses.
If you don't hand in the quest, you cannot access his level.

It used to be until the developers realized that people were totally abusing duncan with farming.. so they enforced the "all bosses" deal before you could go for duncan.. you MUST complete the other areas and bosses before you can go after duncan a second time.
