I searched but didn't find an answer.

Once I dedicate a Destroyer weapon can I continue to use it or is it "lost" to the HoM?

Also, if the weapon is dedicated un-inscribed can I later add/change an inscription or otherwise mod the weapon?

Dedicating it simply customises it,you can still use it but only on that character (and on his/her heroes).

Yes. You can continue to use whichever weapon you decide to dedicate in the Hall of Monuments. However, it will then be customized; you will be unable to sell it or use it on any other character than your own. The character who customized it can also have his/her heroes use it.

Yes. You can mod the weapon before or after dedication however much you would like. There is no restriction to modding it after dedicating.

Destroyer weapons are just like other gold inscribable weapons. Dedicating them in the hall automatically customizes them, but the physical item is still yours. A that point, only you and the heroes attached to that character may use them. Interestingly, the HoM is the only way to "customize" a shield to a character, other than the shields from the bonus mission pack and the /bonus items.
