yes it's possible
i blast through the missions to get ported to the next town asap, and if i feel like doing the skill quests, i do those.
missions have to be done, but quests do not.

Just remember that you hardly get experience without quests/missions. So its not entirely possible to do it without quests: you need to level now and than or it will become too hard to continue!

the only one that I know of that is definitely required is the Academy mission that gets you out of the pre-searing.

I think you need to complete another one . . . sanctum clay or something like that to cross the ocean to amnoon oasis, but I suppose you wouldn't have to do every mission

you cannot get to the crystal desert to ascend unless you do the mission to get there then you cannot ascend unless you do 3 missions then you cannot get to the fire islands or the tomb of primeval kings unless you complete the missions that take you there.
you can skip a whole bunch if you wanted to, but imo it's a hell of a lot easier to get from one place to the next by doing missions. You also get 2k xp and a skill point per mission, so i'd say you'd have to be either a glutton for punnishment or pretty stupid to not do the missions.
