So I have a Tyrian Ne/Mo who hasn't Ascended yet, but has gotten to Senji the Profession-Changer in Cantha.
Presumably, if I do the Mesmer's Path, the Elementalist's Path, and so on, I'll get some free skills in each new secondary. And if I give Senji his 500 gold to change professions, I'll get some free skills.
Are the skillsets identical?

No, they are not.
I had already completed The Mesmer's Path in Tyria long before Factions was around, but I don't normally like that class combination, so I didn't buy anymore skills. When I randomly changed my class to R/Me in Cantha, I unlocked a Mesmer skill that I didn't have before, and had never had. I was a little shocked, but I got a free skill unlock, and probably a few more.
So, it would actually be a good idea for players seeking a universal unlock on a specific character to do both, and change profesions at both places.

I also read in another thread that if you unlock at Senji's, you can't automatically change profession in Tyria. This suggests strongly that it doesn't matter what order you open up each secondary in the two possible ways.
