I just did The Great Northen Wall mission. But no matter what I can't seem to get the mission. Is it possible to get it?
Can cathan born characters do Cities of Ascalon quest?

Why not? I would think that all quests/missions would be available. How do you know this for sure? Inquiring minds want to know.

Missions are doable but Quest aren't. This way you can get 100% Explored.
You will see Quest start appearing in Kryta.

It's the same reason you can't do quests in Shing Jea Monastery as Tyrian characters. Try talking to Master Togo at Shing Jea, and he will tell you that foreigners aren't allowed to study at the monastery.

This was prior to the emperors declaration not to let foreigners study there.

Storyline and NPC dialogues are still not perfect perhaps and will definately change but fact is...
- you can do all of the missions (you can do defend the wall aye) regardless whether you are Tyrian or Canthan born.
- and you can do all the quests which are related to the 1st city you appear in the new continent and onwards (Kryta and Kaineng)
- you can do a few of the quests important for the missions (Welcome to Kryta triggered by Aidan) as well but that is it
- you can not do any skill quests before these starting points.
The motivation? One of my guesses would be to have a different startup skill lists between Tyrian and Canthan chars. Especially if you consider that some of the skills are simply renamed, if you want to use this lovely skill twice in your bar... you have to pay for it
