If I take my Canthan to Lion's Arch is it then possible to go "backwards" to Ascalon?

Yes, you can. BUT - you won't be able to do any of the quest that are available to Tyrian born characters in areas before LA. You will, however, be able to do all of the missions & bonuses.

Yes, but Anet has turned off the rewards for doing missions, see the update notes on guildwars.com.

I doubt there are any quests there for you also, but haven't made the journey myself.
