How do you ascend? Complete the 3 missions in the Crystal Desert and kill your doppleganger. Once in Droknar take the quest from Vanyi.

How to get run? Beacon -> Droknar like always. I'll assume Vanyi is ignorant, like most NPCs, and gives the quest whether or not you've ascended.

like he says pick up the quest from vanyi (heroes journey i think????) and speak to the npc at destinys gorge agostos the brave or something like that.

After you complete the quest from Vanyi, you can take the new quests from the NPCs in each of the three outposts. You should be able to take the quest WHILE doing the quest from Vanyi, but maybe it's been too long for me to remember correctly.

If you can't unlock the quest from Vanyi, go back and ascend properly (if you were run).

Please leave more detailed questions in your posts. I'm rather tired of trying to answer multiple questions that I have to think up myself.
