Does an item like Giantslaying mod over ride anything? And if so, what exactly on what weapons?
I've been considered killing some giants and stuff lately and I am interested in modding a weapon or two.
What does an item like giantslaying override?

It overrides the suffix of the weapon... whatever came after the weapon name, like Enchanting in "Vampiric Long Sword of Enchanting", will get replaced.

Why would you want a weapon that is so limited? Only to farm one single species?

Giants are rather rare, so a weapon "of Giantslaying" will be limited to a very small part of the game. IMHO the +20% damage aren't worth it. You'd be better off using a suffix that has general use, for example a suffix "of Defense".
If you absolutely want a suffix of ____slaying, then use a suffix of Deathbane (Undeadslaying). Deathbane is very useful in UW/FoW. Or Dwarfslaying, which is very useful in GF/SF.

The extra damage can be useful if you're farming one type of monster. A Giantslaying mod can give up to 20% extra damage to giants. Thats an significant amount of extra damage. Since a lot of people carry at lest 2 weapons and switch as necessary, having a weapon geared for farming has its uses.
