No. You can still "unlock" those parts of the map by walking around them.

If they are not "core" or "Factions-only" skills as listed on Guildwiki, then no.

Remember that enemies do not spawn until you kill the boss that comes before them. Always, ALWAYS kill the bosses only when you have nothing else...
