As long as you have it equipped when you cast the enchantment, it will have the 20% bonus.

I wanted to put up a strong build; but nothing too standard. I decided a Blessed Light monk would be a strong premade....

I have never seen a Reduced Blindness single-handed weapon, but if there is such a thing then yes; it should stack.
